Adipocyte 11ßHSD-1 Activity
Glucocorticoid excess is associated with visceral obesity and insulin resistance. Cortisol, a bioactive glucocorticoid, can be produced in adipocytes by 11ß-hydroxysteroid dehydrogenase (11ß-HSD) conversion of cortisone. Inhibition of this activity may be an effective treatment for obesity and metabolic disease. We assay the production of cortisol by human adipocytes in a 96-well format. The assay can identify activators or inhibitors of 11ß-HSD-1 activity.

Figure 1 shows the activation of cortisol production by increasing TNFα concentrations.

Figure 2 shows the inhibition of TNFα stimulated cortisol production by glycyrrhetic acid.
