PCT Media: CnT-17, CnT-15
Through specific formulations designed to target and retain the progenitor cell population in the starting tissue samples, these fully defined media offer significant improvements in isolation and growth, and can even equal or outperform non-defined alternative formulations.
Seeding density is an important consideration when isolation and culturing airway epithelial cells. For PCT media, the optimum density has been found to be in the range of 4x10^3 cells per square centimeter.
As a result of the PCT formulation, differentiation of cells in PCT media has been found to be delayed in certain situations. For any experiments in which cells are induced to differentiate, cultures should be switched to the non-PCT equivalent medium 24-hours prior to the induction of differentiation.
Non-PCT media: CnT-23, CnT-35
These non-PCT media should be used for any differentiation experiments. They do not contain the PCT factors, but do contain retinoic acid, and are low in calcium.
Cells isolated and maintained in a PCT medium should be switched to the equivalent non-PCT medium 24-hours before differentiation is induced by the addition of 1 mM calcium (Ca2+) or your normal protocol.
