Fatty Acid Uptake Assay

Diabetes is a prevalent metabolic disease affecting more than 25 million Americans. About 90% to 95% of patients have non-insulin dependent diabetes mellitus (NIDDM) or type II diabetes. One of the hallmarks of type II diabetes is decreased sensitivity of muscle and adipose cells to insulin. Compounds that increase the sensitivity of adipose to insulin may be useful in the treatment of diabetes and its complications.

Fatty acid uptake assays are performed in 96-well format using mature human adipocytes. Insulin stimulates fluorescently labeled fatty acid uptake into the cells, which is quantified by fluorimetry. Assays can be formatted to investigate the stimulation, inhibition, or sensitization of adipocyte fatty acid uptake.

Positive control: 100 nM insulin
Negative control: TNFα (5ng/ml)

Fig. 1 shows BODIPY-fatty acid uptake in human subcutaneous adipocytes in response to increasing insulin concentrations.
