Rodent (3T3-L1) Lipid Accumulation Assays

Lipid accumulation assays provide the tools to study the compounds that stimulate cultured adipocyte differentiation or lipogenesis. Such compounds are potentially useful in the treatment of diabetes and metabolic disease.

Lipid Accumulation

Our lipid accumulation assay examines the ability of compounds to stimulate lipogenesis during the differentiation of preadipocytes to adipocytes. Cultured preadipocytes are incubated in the presence of appropriate initiation medium, and test compound. Under these culture conditions and in the presence of the test compound there should be a substantial increase in the amount of lipid accumulation when compared to a vehicle control. The differentiation conditions can be customized to suit the contracting scientist.s requirements.

Positive control: Differentiation medium containing PPAR Gamma agonist (10μM) and Dexamethasone (1μM) Negative control: Differentiation medium + TNFa (5ng/ml)

Inhibition of Lipid Accumulation

Our inhibition of lipid accumulation assays examine the ability of compounds to inhibit lipid accumulation in 3T3-L1 cells that are in the process of differentiating to adipocytes. Cultured 3T3-L1 cells are incubated in the presence of differentiation medium and test compound. Under these culture conditions and in the presence of an effective test compound there should be a substantial decrease in the amount of lipid accumulation when compared to controls. Positive control: Differentiation medium + TNFa (5ng/ml)
Negative control: Differentiation medium containing PPAR Gamma agonist (10μM) and Dexamethasone (1μM)

The total triglyceride assay involves complete lysis of the cells. Triglyceride is converted to glycerol and free fatty acids, and the glycerol concentration is measured.

Fig. 1 shows accumulation of triglycerides in 2 week old 3T3-L1 adipocytes. Cells were exposed to a DM-2 differentiation medium, DM-2 plus TNF alpha, or 0.1% DMSO vehicle control. TNF alpha inhibits triglyceride accumulation, whereas complete differentiation medium stimulates triglyceride accumulation.
